Exposure of red blood cells to ozone resulted in K+ leakage, lipid peroxidation and inhibition of some membrane-associated enzymes. On the other hand, carrier-mediated transport of glucose, leucine, sulfate and glycerol and the nonspecific permeation of glycerol, L-glucose and erythritol were not affected by ozone. The cellular level of reduced glutathione declined, whereas the ATP content of the cells was quite insensitive to ozone exposure. It was shown that, most probably, lipid peroxidation and K+ leakage are not causally related. Further, K+ leakage did not reflect gradual, progressive loss of K+ from all cells simultaneously, but occurred in an all-or-none fashion. Finally, ozone-induced damage was compared to damage induced by H2O2, t-butyl hydroperoxide and photosensitizers plus light. It appeared that the pathways leading to membrane deterioration are quite dissimilar in these various forms of oxidative stress.  相似文献

This mainly methodological paper describes the novel use ofan established EEG-based brain imaging technique (Brain ElectricalActivity Mapping or BEAM) to explore human infant responsesto odours. The aim of this study was to evaluate BEAM as aninvestigative tool in infant olfaction. A small sample of 3-month-oldinfants was tested in a low ambient odour environment. The infants'cortical responses to a number of proprietary baby-food odourswere recorded. Two experimental hypotheses were addressed. Thefirst concerned relative differences in cortical response beforeand during odour presentation. Human aortic endothelial cells (HAEC) and human umbilical vein endothelial cells (HUVEC) were labeled with 35SO(4)2- for 48 h. The membrane-associated proteoglycans were solubilized from these monolayers with detergent and purified by ion-exchange chromatography on Mono Q, incorporation in liposomes, and gel filtration. The liposome-intercalated proteoglycans were 125I-iodinated and treated with heparitinase before SDS-polyacrylamide gel electrophoresis. Radio-labeled proteins with apparent molecular masses of 130, 60, 46, 35, and 30 kDa (HAEC) and 180, 130, 62, 43, and 35 kDa (HUVEC) were detected by autoradiography. Further characterization by affinity chromatography on immobilized monoclonal antibodies and by Northern blot analysis provided evidence for the expression of syndecan, glypican, and fibroglycan in human endothelial cells. Most of the heparan sulfate which accumulated in the subendothelial matrix was implanted on a 400-kDa core protein. This protein was immunologically related to perlecan and bound to fibronectin. Binding studies on immobilized antithrombin III suggested that all membrane-associated heparan sulfate proteoglycan forms had the capacity to bind to antithrombin III but that high affinity binding was more typical for glypican. Most of the proteoglycans isolated from the extracellular matrix also bound only with low affinity to antithrombin III. These results imply that glypican may specifically contribute to the antithrombotic properties of the vascular wall.  相似文献

The coding sequence of the uvrA gene from Escherichia coli has been fused to the early promoter, enhancer and origin of replication of the simian virus SV40, and was supplemented with splicing and polyadenylation sites arising from the same virus. Introduction of this hybrid gene into simian cos-1 cells results in the synthesis of a full length UvrA protein (114 kD) which has retained its ability to bind to single-stranded DNA.  相似文献

The vitamin D binding protein (Gc) and posttransferrin-2 (Ptf-2) phenotypes have been determined in a number of Belgian cattle breeds. A very slow migrating variant of the Gc protein — Gc C — has been found in White and Red East Flemish breed. This variant was absent from the other breeds studied. This slow variant was identified as a vitamin D binding protein by autoradiography. The Gc C protein was shown to be controlled by a codominant autosomal allele Gc C at the Gclocus. The Gc C protein is probably identical with a fraction previously described in buffalo and an Italian cattle breed. The allele frequencies for the Gc and Pft-2 systems are reported for several Belgian breeds of cattle.  相似文献

The present investigation is concerned with the use of classical and nonparametric techniques in the analysis of experiments. Occasionally biometricians are confronted with results arising from an experiment with data which do not necessarily satisfy the assumption of Normality, the basis of the classical analysis of variance methods. Sometimes the biometrician handles such a situation by transformation of the observations and applying the classical techniques. Although this attack may lead to a satisfactory analysis, the results may be questionable in a number of cases. In analyzing continuous data without the Normality assumption nonparametric methods provide realistic alternatives. In this paper a number of problems will be discussed with and without the Normality assumption resulting in the well-known classical analysis and its nonparametric counterparts. Most of the nonparametric procedures to be discussed are based on ranks.  相似文献

Summary In this paper the leaf-marking method as used for the study of the development and initial decomposition of floating leaves is described and the reliability of the various measurements is tested and/or discussed. Some general results obtained withNymphoides peltata (Gmel.) O. Kuntze in tanks and in the field are presented and crltically discussed. Autolysis followed by microbial decay was in all cases the most important factor by which leaves disintegrated. In the field plots animals were responsible for the disappearance of 22% of the total leaf area produced during a growth season. This is, however, the combined effect of consumption and damage succeeded by microbial decay. Real grazing can be estimated to be no more than 10% of the production of floating leaves. Fungi can have an important role in initial decomposition, especially after the flowering period, as is demonstrated forSeptoria villarsiae Desm. All damage types show temporal and, in the case of animals, also spatial distribution patterns.  相似文献

A gas chromatographic method is reported for the quantitative analysis of two metabolites of mefruside, viz., 5-oxo-mefruside (mefruside lactone) and its hydroxycarboxylic acid analogue in human body fluids. Use was made of extractive methylation as the derivatization technique, and quantitation was achieved, with a suitable internal standard, by means of a nitrogen-sensitive detector.Because the two metabolites are linked chemically through a lactone—open acid equilibrium, interconversion prior to their separation had to be avoided. A pH partitioning study was performed to find optimal separation conditions. The lactone could be extracted quantitatively at pH 7.4, without any trace of co-extracted hydroxy acid. The latter was extracted either at pH 2 directly (in the case of plasma and urine), or after conversion to the lactone at pH 7.4 (in the case of red cells or whole blood). Concentrations down to 25 ng per sample of both compounds could be analysed with a standard deviation of 5%.The two metabolites of mefruside equilibrated instantaneously between red cells and plasma in vitro. At 37°, the red cell/plasma concentration ratio was 20 for the lactone, but only 0.1 for the open acid compound. 5-Oxo-mefruside was able to displace mefruside from its red blood cell binding sites in vitro.  相似文献
